The 325th prohibition is that we are forbidden to perform melachah on Shavuos.
The source of this commandment is G‑d's statement, "No servile1 melachah may be done [on that day]."

This phrase, m'leches avodah, excludes those types of melachah involving food preparation. Nevertheless, there are many laws regarding the conditions necessary in order to be able to perform such a melachah. See the Shulchan Aruch for the relevant details.
